Oxidation of Phospholipids by OH Radical Coordinated to Copper Amyloid-β Peptide—A Density Functional Theory Modeling

Oxidative stress and metal dyshomeostasis are considered crucial factors in the pathogenesis of Alzheimer’s disease (AD). Indeed, transition ions such as Cu(II) can generate reactive oxygen species (ROS) via O2 Fenton-like reduction, catalyzed by coordinated to amyloid-beta (Aβ) peptide. Despite intensive efforts, mechanisms ROS-induced molecular damage remain poorly understood. In present paper, we investigate, on basis Density Functional Theory (DFT) computations, a possible mechanism OH radical propagation toward membrane phospholipid polar head fatty acid chains starting from end-product generation Cu(II)-Aβ. Using phosphatidylcholine model single unit inside membrane, evaluated thermochemistry with oxidation C-H bond formation moiety. The DFT results show that Cu(II)-Aβ-OH oxidize only sn-2 bonds easily adjacent carbon–carbon double mono or bis unsaturated chain. These discussed recent literature vitro Aβ metal-catalyzed implications AD oxidative mechanism.
